RP-HPLC reversed-phase high performance liquid chromatographic method with UV detection was developed and validated for separation and determination of ampicillin in Iraqi healthy volunteers serum.. Mobile phase consisting of a mixture of deionized water acidified with acetic acid (0.1 %) and acetonitrile as ratio (80: 20 v/v) was delivered at a flow rate of 1.0 ml.min-1. The column was Phenomenex C-18 (50 × 4.6 mm I.D) and 3 m particle size. The eluent was monitored by UV detection at 254 nm. Analysis was performed at room temperature (~25 oC) and the total run time was 10 min. Analytical parameters linearity, accuracy, precision and specificity were determined by validation procedure and found to be satisfactory. Calibration curve was linear over the concentration range of 0.02 – 15 μg/ mL with an R2 of (0.9994). The limits of detection (LOD) and quantification (LOQ) were 0.02 and 0.066 μg/ mL, respectively. Overall, the developed method was found to be simple, rapid, precise and accurate for quality control of ampicillin in volunteers serum.
