- ABSTRACT : Aflatoxin B1 (AFB1) is a common contaminant of foods, particularly in the staple diets of many developing countries. To evaluate genotoxicity impact of AFB1 on gene expression of caspase-3A in common carp (Cyprinus carpio). Caspase-3A plays a pivotal role during caspase-3A dependent apoptosis. A total of 300 common carp with average 45±5 g in weight obtained from Mahaweel fish farm in Babil governorate to Animal and Fish Resource Research Center, fish were randomly distributed into 15 tanks, with five experimental groups; (1) control fed with normal diet without solvent and AFB1, (2) positive control received feed with the only solvent, and (3-5) fed on diets containing 0.5, 1 and 2 mg /kg of AFB1, respectively for 12 weeks. In this study, gene expression of caspase-3A was determined in the liver tissue by semi-quantitative RT-PCR, total RNA was extracted with TRIzol then converted to cDNA and the expression of the genes of caspase-3A and â-actin were conducted by using real-time quantitative PCR assay (qRT- PCR). For caspase-3A gene, both ÄCt and 2–ÄCt values were significantly higher in AFB1 treatment groups than in control groups. The highest level of caspase-3A gene expression in T3 group was 104 fold compared with negative controls 1.00 with ratios (104:100) depending on 2–ÄCt method. While these ratios of the gene expression fold, depend on 2–ÄÄCt method, which was (103:1). Results indicated that AFB1 could significantly increase of gene expression to caspase-3A in hepatocytes, which used as a mechanism to eliminate the genotoxicity effect of AFB1 in hepatocytes.
