It is beneficial to PEGylate the Octreotide (OCT), in order to enhance its biological and physiochemical properties, and to give a longer circulation time. OCT was covalently linked to methoxy- polyethylene-glycol with amine terminal (Me-PEG-NH 2 ). First, Me- PEG-NH 2 was succinylated with succinic anhydride, then the reaction was established at low pH to react the Me-PEG-COOH selectively to the N-terminus of OCT. The covalent attachment of OCT to Me-PEG- COOH was confirmed by the increase in molecular weight by MALDI-ToF mass spectroscopy, which yielded the expected molecular weight of Me-PEG-OCT. The integrity of Me-PEG-OCT and its purity were examined using HPLC. The percent conversion to Mono-PEG-OCT was 71%, as determined by 1 H-NMR. The formed PEGylated-OCT may serve as a linker for coating different nanoparticles which forms a hydrophilic protective layer around the nanoparticles, which prevent and block the opsonization process around the nanoparticles.
